THE EXPRESSION OF B7-H3 ISOFORMS IN NEWLY DIAGNOSED GLIOBLASTOMA AND RECURRENCE AND THEIR FUNCTIONAL ROLE Glioblastoma (GBM) is the most frequent primary tumor of the nervous system with a median survival of 16 months, despite the combination of radio- and chemotherapy after surgical resection. This poor prognosis is the consequence of systematic relapses. GBMs are clearly shown to be heterogeneous in the sense that only a restricted population of tumor cells, GBM stem-like cells (GSCs), can give rise to tumor. Using an in vivo model, we demonstrated that some GBM cells seeded in cultures from patients resected tumors (and obtained through an active collaboration with the Neurosurgery department of the CHU), specifically migrate to the subventricular zone (SVZ) after being grafted in the right striatum. SVZ is one of the two neurogenic regions of the adult brain and migrating GBM cells present the GSC functional features. A proteomic analysis of these cells allowed us to identify different membrane markers of GSCs and among them, B7-H3. B7-H3 is classically described as an immune check point, just as PD-L1 (or B7-H1). B7-H3 was indeed found to be a marker of GSCs in SVZ (which makes sense as this region is known to be lacking brain-blood barrier (BBB) and thus, exposed to immune cells arriving through the blood stream) but also of tumoral pericytes around tumoral vessels devoid of BBB. B7-H3 is also expressed at higher levels at recurrence than at diagnosis, using paired samples from patients. In this paper, we specifically analyzed the “intrinsic” roles of B7-H3 in GBM cells biology and demonstrate that B7-H3, besides its hypothetic immune regulatory role, increases the GBM cells resistance to temozolomide, the alkylating agent used drug in GBM treatment.
