GIGA - Annual report 2023

platforms 69 68 platforms Platform GENOMICS The GIGA-Genomics platform provides a diverse range of DNA and RNA sequencing services, from Sanger and high-throughput genotyping to short and long-read sequencing. Next generation sequencing (NGS) has broad applications in research, including spatial and single-cell transcriptomic profiling, and the identification of DNA and RNA secondary modifications. Additionally, highthroughput genotyping is gaining momentum in human medicine, where it is used in polygenic risk score predictions. We now have access to a GridIon and two P2 solo instruments for parallel sequencing. Duplex runs (where both strands of the same molecule are sequenced), generating over 50% duplex reads with a high quality (Q score above 30), are now possible. ONT sequencing is not limited to long reads; it’s also suitable for sequencing medium-sized amplicon fragments (1 to 2 kb). For single-cell transcriptome analysis, we acquired a Chromium iX system, allowing us to analyze fixed cell suspensions from various patient samples simultaneously, optimizing workflow and reducing costs. Additionally, a new high-density genotyping array and a loading robot streamline the human polygenic risk score calculations pipeline.
