platforms 74 platforms 75 Platform Cell Imaging & Flow Cytometry GIGA-Cell imaging & Flow cytometry provides access to advanced light microscopy systems such as wide-field, point scanning confocal, light-sheet, and super resolution. The facility also offers various cytometry services from simple to multiple immunostaining analysis (using conventional or spectral detection) and 4-ways cell sorting. The core facility’s main goal is to assist researchers in utilizing these powerful tools to address their research questions. Left to right : Gaëtan Lefevre, Céline Vanwinge, Raafat Stephan, Sandra Ormenese, Alexandre Hego The flow cytometry part of the platform developed a new configuration on the Cytoflex FACS for the detection of extracellular vesicles. Spectral flow cytometry also continued to be developed, with over 30 panels developed for more than 20 different researchers. The cell imaging part of the platform developed the FLIM (Fluorescence Lifetime Intensity Microscopy) technique on the Leica Stellaris 8 confocal microscope. This enabled the users to separate 3 spectrally identical fluorophores on living cells, based on their lifetime. Multiplexing with 8-color protocols has also been developed, and the platform’s staff plan to couple FLIM with multiplexing experiment to do more than 10 colors at a time. The new multiplexed images have enabled to develop the spatial colocalization analysis technique to highlight the distance between immune cells in a tumor or asthma context. Simultaneously, the Axioscan 7 slide scanner was developed, featuring 7-color protocols, as well as white light and polarized light acquisitions. Lastly, a tutorial about the Cellpose artificial intelligence segmentation tool was written to enable any researcher to set up their computer and use this tool. Cellpose helps biologists to identify cells in images from microscopy. In 2024, the platform will expand its services by developing spatial transcriptomics. The confocal microscope park will be strengthened by the acquisition of a new spinning disk microscope for high-speed 3D live cell imaging.
